Abstract

Preanalytics for lipidomics encompasses a broad range of topics that include everything that occurs with samples before analyzing the lipids by mass spectrometry. Preanalytics begins with experimental design and planning for acquisition of samples. Storage and preservation of these samples play a vital role as the samples may not be analyzed for months or years. Homogenization and extraction of lipids from biological samples are key steps in this process. Understanding the solubility of lipids and how temperature can affect it is critical. Automation can play a key role in increasing the throughput or just increasing the precision and accuracy. Finally, a collection of “tips and tricks” are offered based on our experience doing lipidomics for almost two decades. All these topics, as well as others, are critical because if samples are not collected, stored, and extracted correctly, analysis by mass spectrometry will be futile. Samples often have limited quantities and are not replaceable without tremendous effort; therefore, careful attention to preanalytics is of utmost importance. Careful attention to all aspects of preanalytics will set a solid foundation for analysis by mass spectrometry and be the basis for quality lipidomic data.
