The effects of diphenylhydantoin and spironolactone on the digoxin pharmacokinetics in the rabbit

Abstract

We have studied the effects of diphenylhydantoin and spironolactone pretreatments on the digoxin pharmacokinetics in female albino rabbits using the radioimmunoassay technique. The serum digoxin concentration time curves of the control series, as well as the pretreated series, exhibit a biexponential component according to the equation C = A0 x e-(αt) + B0 x e-(βt). A significant increase in the half lives in both 'fast' and 'slow' phases of the digoxin concentration-time curves was found in the diphenylhydantoin pretreated animals, as well as a slowing in the distribution rate constants and a decrease in the elimination rate constant of the glycoside. The spironolactone induces a significant prolongation in the half life of the 'fast' phase of the digoxin concentration-time curve and a decay in its distribution rate constant from the central to peripheric compartment, as well as an increase of the apparent volume of distribution in the central compartment.
