Duplex RNAs that target gene promoters are one strategy for enhancing the expression of target genes. This chapter summarizes our understanding of mechanism behind gene activation by small RNAs that bind noncoding transcripts at gene promoters. We describe a path for choosing candidate genes for therapeutic development.",
