Pharmacokinetics of cefaclor in infants and children

Abstract

The pharmacokinetics of cefaclor were studied in 28 infants and children. Serum concentrations of 10.8 μg/ml and 13.1 μg/ml were observed in fasting patients 30min after 10 and 15 mg/kg doses, respectively. Although peak concentrations were smaller in children given drug and milk concomitantly, the area-under-the-curve values were not affected. Average half-life values ranged from 36 to 60 min; shorter times were observed with 15 mg/kg doses. The mean concentrations of cefaclor in saliva at 2,4 and 6 h after 15 mg/kg doses were similar to those in serum. The levels in tears were > 1.0 μg/ml in 7 of 8 samples obtained 2 h after the dose. These pharmacokinetic properties of cefaclor were compared to those of cephalexin in comparably aged patients.
