Abstract
A high-pressure liquid chromatographic (HPLC) assay was developed for the determination of the epimeric distribution of ticarcillin and a derivatization method was used for the HPLC assay of clavulanic acid. Fifty-five women in labor who underwent cesarean section for cephalopelvic disproportion were given 3.1 gm of Timentin (ticarcillin 3.0 gm and 100 mg of clavulanic acid) intravenously, and the total serum ticarcillin and clavulanic acid levels measured throughout four hours. The epimeric distribution of ticarcillin, and the ratio of ticarcillin to clavulanic acid was also determined. The mean peak serum concentration was 205.7 ± 89.4 μg/ml at 30 minutes post dose and there were no changes in the epimeric distribution of ticarcillin. The simultaneous mean peak clavulanic acid serum concentration was 3.6 ± 1.6 μg/ml. The half-life of ticarcillin and clavulanic acid was 49.9 ± 16.3 minutes, and 67.9 ± 24.4 minutes respectively. There was a significant difference in these half-lives compared to non-pregnant individuals.
