N2 - A survey of fluorescence characteristics of tetrahydrofolate and related compounds has been carried out with an Aminco-Bowman Spectrophotofluorometer. Dihydrofolate and all the tetrahydrofolate derivatives examined, except 5,10-methenyltetrahydrofolate, show maximum excitation between 300 to 320 mμ and maximum fluorescence between 360 to 425 mμ. On the other hand, folate and the related compounds exhibit an excitation maximum at 360 to 380 mμ and a fluorescence maximum at 450 to 460 mμ. The fluorescence intensity of tetrahydrofolate increases markedly in acidic solutions. This property provides a simple and a highly sensitive method for a quantitative determination of tetrahydrofolate.
